Offer Description This thesis project will be developed within the framework of the PHOSPHO-gi-RUS project funded by the ANR, which consists of exploring the role of phosphorylation in the viral cycle of Pandoraviruses, by combining biochemical, bioinformatics, phosphoproteomic, structural and imaging approaches, for the spatial, temporal and structural characterization of the identified kinases.
Pandoraviruses are a family of giant viruses (girus) that infect the protozoan Acanthamoeba. Their genome encodes 500 to 2500 proteins, most of which have few or no cellular and viral homologs. Recently, several proteins with catalytic domains similar to eukaryotic protein kinases have been identified, but their functions in the host infection cycle have not yet been characterized.
The proposed thesis project will aim to develop fluorescent peptide biosensors to characterize the kinase activity of viral proteins identified as potential kinases, thanks to the expertise of the host laboratory which has already developed a toolbox of substrate peptides and reporter biosensors of human kinase activities with several proofs-of-concept in vitro, in cellulo and in vivo (for review Morris M.C. Life 2022; Morris M.C. EurJOC 2022). First, selective biosensors, allowing sensitive and quantitative detection of viral kinase activities, will be developed and characterized. These tools will then be applied to the study of the functions of viral kinases in the viral cycle and in the host infection process in vitro and in vivo. Finally, these biosensors will be used to screen small molecule libraries to identify inhibitors to block certain steps in the viral infection cycle.
