Offer Description Full-field multispectral Mueller polarimetric imaging for improved surgery of neurological malignancies

Problem. Surgery is the crucial step in the treatment of brain tumors, particularly gliomas. While some well-defined tumors can be removed en bloc, the majority of gliomas grow infiltratively in the white matter of the brain and are removed piecemeal. It is essential to identify and respect the boundary between brain tumor and surrounding healthy brain tissue to carry out a radical resection while preserving neurological function. However, solid tumor tissue is often difficult to differentiate from infiltrated white matter during surgery, even with advanced intraoperative microscopes. Non-complete tumor resection due to poor visualization of tumor margins leads to a worse prognosis as tumors grow back from the remnants. Various imaging techniques (e.g., fluorescence imaging, ultrasound, and magnetic resonance imaging) have been implemented for intraoperative visualization, but each has drawbacks. Thus, efforts to visualize brain tumors and reliably identify the interface between healthy and pathological areas during neurosurgery have not been successful for many intrinsic brain tumors.
Approach. Imaging Mueller polarimetry is an optical technique that has been widely used for studying biological tissues, which represent complex depolarizing and optically anisotropic media. This technique is sensitive to microstructural changes in biological tissues induced by pathology. Brain tumors destroy the highly anisotropic structure of healthy white matter, which is made up of dense, well-organized axon fibers. Accurately detecting healthy white matter can help surgeons better identify pathological areas. The optical team of LPICM has developed several Mueller polarimetric imaging systems (e.g., multispectral Mueller Colposcope, Mueller laparoscope) that have shown promise for improving in vivo detection of cancerous lesions and accurately defining resection margins. Previous studies conducted within the SNF Sinergia HORAO project by LPICM, Ecole polytechnique, and Bern University Hospital demonstrated that wide-field Mueller imaging systems can provide detailed maps of axonal fiber orientation in healthy brain and effectively distinguish between healthy and cancerous areas in just a few seconds. A miniaturized Mueller imaging system based on a polarization-sensitive camera will be built for real-time intraoperative use during neurosurgery.
Responsibilities. The main objective of the postdoctoral fellow's work is to develop a wide-field Mueller polarimetric imaging system to improve the detection of surgical margins of neurological cancers in vivo . The post-doctoral fellow will focus on two major complementary parts:
Designing and building a prototype of a miniaturized Mueller polarimetric imager using a polarization-sensitive camera, which includes:
Extensive characterization of the polarization-sensitive camera.
Design of a new optical system for polarization modulation of incident light.
Development of electronic control for image acquisition.
Optimization of the illumination and detection arms for instrument miniaturization.
Adapting the miniaturized system to a mechanical robotic arm for in vivo acquisition in the operating room.
Transferring the developed prototype instrument to the University Hospital of Bern, Switzerland.
Assisting a neurosurgeon in collecting in vivo data, processing polarimetric images, and comparing them to histological analysis results.
Expected Competences: Strong expertise in optics, optical instrumentation, design of polarimetric systems, and signal processing. Strong motivation for teamwork in a multidisciplinary environment with continuous interaction with medical doctors. Basic knowledge of matrix algebra is a plus.
Linguistic Skills: Good knowledge of English.
Informatics Skills: Good knowledge of Matlab, Python, Labview.
